Upon meeting a young boy named Haruta who makes a living as a pickpocket, Gintoki discovers that he is pickpocketing out of a desire to meet his long-lost mother, Hiwa, Yoshiwara's number one courtesan. Gintoki and his friends venture to Yoshiwara to fulfill Haruta's wish, but standing in their way is Housen, the Night King who rules Yoshiwara.

In Edo, where Gintoki and the Yorozuya reside, order is maintained by the Shinsengumi. Toshiro Hijikata, known as the "Demon Vice-Commander," has long led the Shinsengumi with strict discipline. However, after obtaining a cursed demonic sword from a blacksmith, his personality drastically changes due to the curse—turning him into a completely useless good-for-nothing. Justas he’s cornered by a group of rogue samurai in a life-or-death situation, the one who saves this pathetic Hijikata is none other than the Shinsengumi’s tactician, Ito Kamotaro, who has finally returned to Edo after completing a long-awaited mission. Kamotaro comes back bearing the latest weapons and prized swords as souvenirs, receiving a grand welcome from Commander Kondo. His accomplishments are highly praised, and he gradually begins rising in prominence within the Shinsengumi.

Kabukicho's delicate balance is maintained by four powerful figures known as the "Four Devas," who keep each other's influence in check. Among them is Otose, the "Empress" of the district. Despite having no real power herself, the sole reason she retains her position as one of the Four Devas and can contend with rival factions... is the presence of Kabukicho's strongest man—Sakata Gintoki—by her side. Gintoki is given anultimatum: Join the Jirochou Family to save her, or refuse and watch Otose be attacked—while hehimself is marked for death.

Having been outlawed for its strong potency, the love drug was thought to have disappeared from the red-light district for good; however, it seems to have found its way back inside Yoshiwara's walls and now has the entire district under its influence. Of those affected is the cold-hearted Hyakka captain, Tsukuyo, and the first person she sees after inhaling the smoke is none other than the brazen Yorozuya leader, Gintoki Sakata. Unbeknownst to everyone else though, she was already attracted to Gintoki. With those hidden feelings now intensified, Tsukuyo must overcome her afflictions and work with the Yorozuya to return everyone back to normal.

Saved for the time being, Gintoki enters the Yorozuya office, but unbeknownst to him, someone else is already waiting there... Meanwhile, Kondou departs Earth to marry Princess Bubbles in an attempt to improve diplomatic relations. After boarding the Gorilla Amanto mother ship, he realizes that he doesn't speak their language. Confused, Kondou tries conversing with them, only to inadvertently gain their support. However, someone associated with the princess crashes the ongoing ceremony. Will the wedding continue, or has Kondou just been saved from becoming the next Gorilla Prince?
